The wheels are set to start rolling on Friday, when the first bus leaves Tambar Springs bound for Tamworth, stopping in at Premer, Spring Ridge, Caroona, Walhollow, Werris Creek, Currabubula and Duri on the way.
The route will be trialled for six months.
Tamworth business Fiona's Minibuses will run the service, and the bus will leave Tambar Springs on Friday mornings and depart Tamworth on Friday evenings.
The state government announced the new route earlier this week.

Member for Tamworth Kevin Anderson said the public transport option will help connect outlying towns to the city.
"I'm excited to welcome this service because it will give our more isolated communities greater access and connection to vital services in larger centres," he said.
"An extra bonus with the service is that passengers can also connect at Tamworth with NSW TrainLink services to Sydney.
"For some of these communities, this will be the first time they've had access to a public transport service and will really change the way many people move around the district."
The bus is scheduled to depart Tambar Springs at 7:30am on Fridays, and it will leave the first Tamworth stop at 3:50pm on the same day. The route is just under two-and-a-half hours.
The full timetable, including three Tamworth stops, can be viewed on the Transport for NSW website, but bookings must be made directly through Fiona's Mini Buses by calling 6760 9084.
Minister for Regional Transport and Roads Paul Toole announced a total of six new regional bus service trials, including Goondiwindi to Inverell.
Transport for NSW will monitor community feedback throughout the six-month trial.
