Molloy, who also hosts Channel 7's The Front Bar, is moving over to the Melbourne drive slot with Jane Kennedy before their show goes national in 2018.
It was speculated that Lawrence Mooney would replace Molloy, but on Friday Hot Breakfast host Eddie McGuire revealed Anderson would be joining himself and Luke Darcy behind the mic.
Triple M's breakfast show is among the most-listened to programs on the FM network in Melbourne, and is popular with men and AFL fans. Molloy's switch will see him go head-to-head with big names such as Dave Hughes and whoever replaces Hamish and Andy.
While making the on-air announcement, Anderson said he was honoured to take up the role.
"[I have] huge shoes to fill, even if those shoes only take 43 steps in a day," he said. "I'm going to aim for 44 steps. The fact they said you can come in at 7am [instead of 6am] really got me across the line."
In turn, Molloy joked that he didn't think Anderson was the right man for the job until he heard about his run-in with authorities while on a flight to Wagga Wagga back in June.
In a statement, Anderson said he's always been a "huge fan" of Triple M's breakfast show.
"It's been my lifeline to Melbourne as I have toured the world doing stand-up," he said. "I'm guessing I've probably heard more of it than Mick has."
The comedian and Gruen Transfer host said the show will put his skills to good use.
"It's a show that gives you the news, and makes the news, all with a laugh," he said. "So many times I have thought I would love to be part of that conversation, and now I'm rapt that I get to, waking up with my favourite city in the world."
Anderson, who has been filming the latest series of Gruen, has confirmed he will still have time to fulfil his ABC commitments while juggling breakfast radio.
